The Assay Genie CXCL10 ELISA Kit enables quantitative measurement of interferon-gamma-inducible protein 10 (IP-10/CXCL10) in bovine serum and plasma samples via a sandwich ELISA workflow on a 96-well microplate. CXCL10 is a pro-inflammatory chemokine secreted in response to IFN-γ stimulation, recruiting CXCR3-expressing Th1 lymphocytes, NK cells, and macrophages to sites of infection and inflammation, making it a useful biomarker for veterinary immunology, infectious disease research, and vaccine response studies in cattle.

Reconstituted standards and samples should be handled carefully and stored according to the kit insert; unopened kits are commonly shipped refrigerated and stored at 2-8 °C, with longer-term storage of individual components at -20 °C to preserve activity, and repeated freeze-thaw cycles avoided. Colorimetric detection is typically read at 450 nm using a standard microplate reader, with intra-assay and inter-assay precision generally below 10% CV.

Intended for research use only and in vitro laboratory applications, the kit supports studies of bovine respiratory disease, mastitis, Johne's disease, viral infections such as BVDV, and reproductive immunology. Each kit includes pre-coated wells, detection antibody, standards, buffers, and substrate solution sufficient for 96 assays.

The assay workflow pairs a biotinylated detection antibody with streptavidin-HRP and TMB substrate; the enzymatic reaction is terminated with a sulfuric acid stop solution before absorbance is recorded at 450 nm on a standard microplate reader. Total run time is approximately 2.5-3 hours after sample and reagent preparation, with multiple wash steps between incubation periods. A recombinant bovine CXCL10 standard is included to generate a standard curve used to interpolate unknown sample concentrations across a working range that typically spans low pg/mL to low ng/mL values.

For bovine serum and plasma, an initial 2-fold dilution in the supplied assay diluent is commonly recommended, with spiked recovery values usually falling within 80-120% when samples are kept refrigerated and freeze-thaw cycles are minimized. Individual kit components remain stable when stored at 2-8 °C, with reconstituted standards and detection antibody aliquoted and held at -20 °C for longer-term use. Cross-reactivity screening has shown this type of assay does not significantly cross-react with related bovine cytokines such as IFN-alpha, IFN-gamma, IL-1alpha, IL-13, TNF-alpha, MIG, or MIP-1beta, supporting its specificity for CXCL10. Researchers investigating Mycobacterium avium paratuberculosis infection, bovine viral diarrhea virus, and bovine respiratory syncytial virus have reported elevated IP-10 concentrations as an early indicator of Th1-driven cell-mediated immune activation in cattle.
