Description
Designed for the quantitative measurement of C-C chemokine receptor type 7 (CCR7) in bovine samples, this Sandwich ELISA kit from Assay Genie uses a colorimetric detection method and is compatible with routine laboratory and high-throughput screening formats. The microplate provided is pre-coated with an antibody specific to CCR7. Standards or samples are added to the appropriate wells and combined with the specific antibody, followed sequentially by a biotinylated detection antibody specific for CCR7 and an Avidin-Horseradish Peroxidase (HRP) conjugate. After incubation and washing, a substrate solution is added; the enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution, and the optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of CCR7 in the sample.
The assay accepts a broad range of sample types including serum, plasma, blood, saliva, urine, cell culture supernatant, tissue samples, and other related supernatants. Detailed sample preparation guidance covers serum (serum separator tube, 30 minutes clotting, centrifugation for 15 minutes at approximately 1000 × g), plasma (EDTA or heparin as anticoagulant, centrifugation for 15 minutes at 1000 × g at 2°C–8°C within 30 minutes of collection), urine (centrifugation for 20 minutes at 2000–3000 rpm), and cell culture supernatant (centrifugation at 4°C for 20 minutes at 2000–3000 rpm). Notes flag that hemolyzed samples cannot be detected, that samples containing NaN3 interfere with HRP, and that samples intended for use within 5 days may be stored at 2–8°C, otherwise at -20°C (1 month) or -80°C (2 months) to avoid loss of bioactivity and contamination.
On receipt, the Assay Plate, Standard, Detection Reagent A and Detection Reagent B should be stored at -20°C and the Substrate at 4°C, with other reagents stored according to the labels. Once opened, the kit remains stable until the expiration date, which is six months from the date of manufacture, provided it is stored as prescribed. The product is intended for research use only.
