BLNK Colorimetric Cell-Based ELISA Kit

Assay Genie's BLNK Colorimetric Cell-Based ELISA Kit provides a convenient, lysate-free, high throughput and sensitive assay for detecting B-cell linker protein (BLNK) expression profiles in cultured cells. The cell-based format allows researchers to measure relative levels of BLNK in cells and to screen the effects of treatments, inhibitors such as siRNA or chemicals, and activators on BLNK expression.

The kit uses an indirect ELISA format: BLNK is captured by BLNK-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The bound HRP enzyme catalyses a colorimetric reaction on substrate addition, read at 450 nm. The assay is reactive with human and mouse samples, is intended for use with adherent cells, and supports a dynamic range greater than 5000 cells. Storage is recommended at 4 °C with a stability of 6 months.

For normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control. Following the colorimetric HRP measurement, the Crystal Violet whole-cell staining method can be applied to assess cell density, and absorbance values can be normalised to cell numbers to correct for plating differences.
