Description

From Assay Genie, this Cell-Based ELISA Kit enables the detection of phosphorylated tyrosine 807 on the BL-CAM (Basal Cell Adhesion Molecule) protein in cell lysates and tissue extracts. It uses a colorimetric detection method at 450 nm and is suitable for adherent cell samples, offering a lysate-free, high-throughput workflow for measuring BL-CAM phosphorylation and expression in cultured cells.

The assay employs an indirect ELISA format in which BL-CAM (Phospho-Tyr807) is captured by phospho-specific primary antibodies, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, while Crystal Violet whole-cell staining can be used to adjust for plating differences by normalizing to cell density. The kit is reactive with human samples and has a stated dynamic range of > 5000 Cells. It can be applied to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on BL-CAM phosphorylation.

BL-CAM, also known as CD22, is a cell adhesion molecule involved in B-cell interactions, migration, and intracellular signaling, with phosphorylation at tyrosine 807 known to regulate its function and downstream signaling cascades. The kit supports research into cell signaling pathways relevant to cancer cell metastasis, immune cell activation, and developmental processes. The associated UniProt entry is P20273, with the gene located on chromosome 19q13.1 and a protein molecular weight of 75,354 Da. The research area is cell biology.
