Description

Assay Genie offers a cell-based ELISA kit for the detection and quantification of BIK protein phosphorylated at Threonine 33 in adherent cells and other biological samples. The assay uses an indirect ELISA format with HRP-conjugated secondary antibodies and a colorimetric readout at 450 nm, providing a lysate-free, high-throughput workflow for measuring relative phosphorylation levels in cultured cells.

Qualitative determination of BIK (Phospho-Thr33) is achieved by capturing the target with phospho-specific primary antibodies, followed by enzymatic conversion of substrate by the conjugated HRP. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining can be used to assess cell density. The kit is suited for screening the effects of treatments, inhibitors (e.g., siRNA or chemicals), or activators on BIK phosphorylation, with a dynamic range of > 5000 Cells.

BIK is a pro-apoptotic protein that accelerates programmed cell death and contains one Bcl-2 homology 3 (BH3) domain; its activity is suppressed by binding to the apoptosis repressors Bcl-xL, Bcl-2, or BHRF1. It does not interact with Bax. The corresponding human gene (Gene ID: 638, UniProt ID: Q13323) is located at chromosomal position 22q13.31, and the encoded protein has a molecular weight of 18,016 Da.
