BIK Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit is designed for qualitative measurement of BIK (Bcl-2-interacting killer) protein expression in cultured human cells. The colorimetric assay uses an indirect ELISA format in which BIK-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction read at 450 nm. The kit is lysate-free and supports high-throughput workflows on 96-well microplates, with a stated dynamic range of greater than 5000 cells. Adherent cells are the intended sample type.

The assay is suitable for drug screening, toxicity studies, and general cell-based experiments, and can be used to monitor relative BIK levels in response to treatments, inhibitors such as siRNA or chemicals, or activators. Because the readout is qualitative, two normalisation strategies are provided: a monoclonal anti-GAPDH antibody as an internal positive control, and a Crystal Violet whole-cell staining step to determine cell density and adjust for plating differences.

BIK is a pro-apoptotic mitochondrial integral membrane protein that accelerates programmed cell death through a BH3 domain by binding the apoptosis repressors Bcl-xL, Bcl-2, and BHRF1 (the Epstein-Barr virus homolog of Bcl-2), but does not interact with Bax. The protein is encoded by the BIK gene (Gene ID 638, UniProt ID Q13323) located at chromosomal position 22q13.31, with a molecular weight of 18,016 Da. The kit is stored at 4°C with a stability of 6 months.
