Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for detecting BCL-2 phosphorylated at Ser70 in cell samples. The assay is cell-based, lysate-free, high-throughput and sensitive, allowing researchers to measure the relative amounts of phosphorylated BCL-2 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on BCL-2 phosphorylation. The kit is reactive with mouse and rat samples, supports a dynamic range of > 5000 cells, and uses colorimetric detection at 450 nm.

Qualitative determination of BCL-2 (Phospho-Ser70) is achieved through an indirect ELISA format, in which the target is captured by phospho-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density so that absorbance values can be normalized to cell amounts and plating differences can be adjusted. The kit is intended for use with adherent cells.

BCL-2 is an antiapoptotic member of the Bcl-2 family that regulates cell death by controlling mitochondrial membrane permeability and by inhibiting caspase activity either through preventing cytochrome c release from the mitochondria or by binding to the apoptosis-activating factor (APAF-1). Phosphorylation at this site has been linked to modulation of its anti-apoptotic function, and dysregulation of BCL-2 phosphorylation has been implicated in diseases such as cancer and autoimmune disorders, making it a relevant target for research into cell survival and signalling pathways.
