Description

Assay Genie provides a Cell-Based ELISA Kit for the quantitative detection of BCL-XL protein levels in cell lysates and tissue extracts. The assay runs in a lysate-free, high-throughput, microplate-based workflow that uses an indirect ELISA format: BCL-XL is captured by BCL-XL-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to drive a colorimetric reaction upon substrate addition.

Designed for use with cultured cells, the kit can measure relative BCL-XL amounts and screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Because the Cell-Based ELISA readout is qualitative, two normalization approaches are supplied: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density so that absorbance values can be adjusted for plating differences. Reactivity covers Human, Mouse, and Rat samples, the dynamic range is greater than 5000 cells, absorbance is read at 450 nm, and the kit is stored at 4°C with a stated stability of 6 months.

BCL-XL (gene symbol BCL2L1; UniProt Q07817; Gene ID 598) is an anti-apoptotic member of the BCL-2 family located at the outer mitochondrial membrane, where it regulates VDAC opening, mitochondrial membrane potential, reactive oxygen species production, and cytochrome c release. Dysregulation of BCL-XL has been linked to cancer and neurodegenerative disorders, making its accurate measurement of interest for apoptosis and cell-survival research.
