Description

From Assay Genie, this high throughput, lysate-free cell-based ELISA is designed to detect BCL-2 protein phosphorylation and expression profiles in cells, specifically measuring BCL-2 phosphorylated at Ser70. The indirect-format assay uses target-specific primary antibodies to capture the phosphorylated protein and HRP-conjugated secondary antibodies that bind the Fc region of the primary antibody, catalysing a colorimetric reaction at 450 nm upon substrate addition. It can be used to measure relative amounts of phosphorylated BCL-2 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on BCL-2 phosphorylation.

Validated for use with human samples and adherent cell cultures, the assay has a dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control in normalising the target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, so that absorbance values can be normalised to cell amounts to adjust for plating differences.

BCL-2 is an antiapoptotic member of the Bcl-2 family that regulates cell death by controlling mitochondrial membrane permeability. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria or by binding to the apoptosis-activating factor (APAF-1). Phosphorylation at Ser70 has been linked to its anti-apoptotic function, and studying this modification can provide valuable insights into cell survival pathways and potential therapeutic targets for conditions such as cancer and autoimmune diseases.
