Description

From Assay Genie, this Cell-Based ELISA Kit enables the accurate detection of phosphorylation levels of the BAD protein at the Ser91-128 site in cell lysates. It uses a colorimetric detection method measured at 450 nm and is validated for use with Human, Mouse, and Rat samples. The assay supports a dynamic range of greater than 5000 cells, making it suitable for high-throughput studies of cell signalling pathways involved in survival and apoptosis.

The kit employs an indirect ELISA format in which BAD (Phospho-Ser91/128) is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colour reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, while a Crystal Violet whole-cell staining step allows cell density to be determined, so plating differences can be adjusted. The procedure is designed for adherent cell samples and can be used to measure relative BAD phosphorylation or to screen the effects of treatments, inhibitors, or activators on the pathway.

BAD is a pro-apoptotic member of the Bcl-2 family, and phosphorylation at Ser91-128 regulates its pro-apoptotic function, linking the kit to research into apoptosis, cell survival, and potential therapeutic targets in cancer and neurodegenerative disorders. The target corresponds to UniProt ID Q92934 (Gene ID 572, OMIM 603167). For optimal performance, the kit should be stored at 4°C with a stated shelf life of 6 months, and standard delivery is 7 to 10 days.
