B-RAF (Phospho-Ser602)Fluorometric Cell-Based ELISA Kit

From Assay Genie, this Fluorometric Cell-Based Phospho ELISA Kit is designed to monitor B-RAF (Phospho-Ser602) protein phosphorylation and expression profile in cells. The kit offers a convenient, lysate-free, high-throughput and sensitive assay format that allows qualitative determination of phosphorylated B-RAF using an indirect ELISA approach, where target-specific primary antibodies are detected with dye-conjugated secondary antibodies that emit at defined wavelengths.

The kit is intended for measuring relative amounts of phosphorylated B-RAF (Phospho-Ser602) in cultured cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on B-RAF phosphorylation. Reactivity is established for Human, Mouse, and Rat samples. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target RFU values, and an antibody against the non-phosphorylated counterpart of B-RAF is also provided for normalisation. Plates are read at Ex/Em: 651/667 (Dye 1) and 495/521 (Dye 2).

The dynamic range is greater than 5000 cells, with recommended storage at 4°C for 6 months. The target B-RAF is a serine/threonine protein kinase of the RAF family with a molecular weight of 84kDa, involved in the transduction of mitogenic signals from the cell membrane to the nucleus and in the MAP kinase/ERKs signaling pathway, which affects cell division, differentiation, and secretion.
