Description

From Assay Genie, this Cell-Based ELISA kit is designed for the qualitative measurement of B-Myb protein phosphorylation and expression in cultured cells, specifically detecting phosphorylation at serine residues 577-581. B-MYB is a transcription factor involved in cell cycle regulation, cell proliferation, and differentiation, and phosphorylation at these residues plays a role in its activation and function, supporting research into cell cycle control and cancer progression.

The assay uses an indirect ELISA format in which B-Myb (Phospho-Ser577/581) is captured by phospho-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The kit is reactive with Human, Mouse, and Rat samples and operates with a dynamic range of > 5000 Cells, with read-out at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control in normalizing the target absorbance values, and the Crystal Violet whole-cell staining method is provided for determining cell density so that absorbance values can be normalized to cell amounts and plating differences adjusted.

Designed as a lysate-free, high-throughput workflow for adherent cells, the kit supports measuring relative amounts of phosphorylated B-Myb in cultured cells as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on B-Myb phosphorylation. The target corresponds to gene symbol MYBL2 (Gene ID: 4605, UniProt ID: P10244), and the kit is classified under the Epigenetics and Nuclear Signaling research area with a Phospho sub-type.
