Description

This Cell-Based ELISA kit from Assay Genie is designed for the qualitative measurement of Aurora kinase A phosphorylated at threonine 288 (AurA Phospho-Thr288) in cultured cells. Using a lysate-free, indirect ELISA format, the assay captures AurA (Phospho-Thr288) with target-specific primary antibodies, and detection is achieved through HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. The kit is reactive with human, mouse, and rat samples, is suited for adherent cells, and offers a dynamic range of > 5000 cells.

Researchers can use the kit to measure relative levels of phosphorylated AurA in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on AurA phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step may be used to determine cell density after the colorimetric measurement.

Aurora kinase A (gene symbol AURKA) is a cell cycle-regulated serine/threonine kinase of the AUR family, localized to mitotic centrosomes and spindle microtubules. It is required for centrosome maturation, centrosome duplication, and chromosome segregation, and is overexpressed in many tumor cell lines. Known inhibitors of the kinase include VX-680, ZM447439, Hesperadin, and SNS-595, providing context for downstream studies of this signalling target.
