AurA Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed for the in-cell detection of AurA (Aurora kinase A; gene symbol AURKA) using a colorimetric readout at 450 nm. The assay targets adherent cell samples and has been validated for reactivity with human, mouse, and rat specimens, with a stated dynamic range of > 5000 cells. The kit is supplied in a 96-Well Microplate format and is stored at 4 °C for up to 6 months.

Working as a lysate-free, high-throughput indirect ELISA, the assay uses AurA-specific primary antibodies paired with HRP-conjugated secondary antibodies, so that the conjugated enzyme drives a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell stain is provided so that cell density can be measured and plating differences adjusted. The same plate format can be used to compare relative AurA levels across treatments, inhibitors (such as siRNA or chemicals), or activators.

Aurora kinase A is a member of the AUR family of cell cycle-regulated serine/threonine kinases that is required for centrosome duplication, chromosome segregation, and mitotic spindle organisation. The protein localises to mitotic centrosomes and spindle microtubules, and is reported to be overexpressed in many tumour cell lines, where it has been associated with transformation and aneuploidy.
