Description

Assay Genie's cell-based ELISA kit is designed for the accurate measurement of phosphorylated ATRIP (ATR Interacting Protein) at Ser68 in cell lysates, supporting research into DNA damage response and cell cycle regulation. Phosphorylation at this site is critical for activation of the ATR (Ataxia Telangiectasia and Rad3-related) kinase pathway, and the assay can be used to measure relative phosphorylation in cultured adherent cells as well as to screen the effects of various treatments, inhibitors, or activators.

The kit uses an indirect ELISA format: a phospho-specific primary antibody captures the target, while an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step is used to determine cell density. The assay shows reactivity with human, mouse, and rat samples, with a dynamic range of > 5000 Cells, and is positioned within Epigenetics and Nuclear Signaling research.

ATRIP is an essential component of the DNA damage checkpoint, binding single-stranded DNA coated with replication protein A and interacting with the ATR kinase to accumulate at intranuclear foci induced by DNA damage. Associated database identifiers include Gene ID 84126, UniProt Q8WXE1, and OMIM 606605.
