Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for the accurate detection of phosphorylated ATRIP at Ser224 in adherent cells. The assay uses an indirect ELISA format, in which target-specific primary antibodies are captured and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalyzing a colorimetric reaction upon substrate addition that is read at 450 nm.

The kit is lysate-free, high-throughput, and sensitive, enabling measurement of the relative amounts of phosphorylated ATRIP in cultured cells as well as screening for the effects of various treatments, inhibitors, or activators on ATRIP phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density for normalization. The dynamic range is > 5000 Cells, and the kit is reactive with human samples.

ATRIP (ATR-interacting protein) is a key component of the DNA damage response pathway, required for checkpoint signaling after DNA damage and important for maintaining genomic stability. Phosphorylation of ATRIP at Ser224 is essential for its function in DNA damage repair, making this modification a valuable marker for studying cellular responses to DNA damage. The kit falls within the Epigenetics and Nuclear Signaling research area.
