ATP-Citrate Lyase (Phospho-Ser454)Colorimetric Cell-Based ELISA Kit

Assay Genie's phospho-specific cell-based ELISA kit is designed to measure ATP-Citrate Lyase phosphorylated at serine 454 directly in cultured adherent cells, without the need for lysate generation. Detection is colorimetric and read at 450 nm, and the assay follows an indirect ELISA format in which a phospho-specific primary antibody captures the target while an HRP-conjugated secondary antibody catalyses the colour reaction upon substrate addition.

Validated for human, mouse and rat samples, the kit has a dynamic range of > 5000 cells. An internal anti-GAPDH antibody is included to normalise target absorbance values, and a Crystal Violet whole-cell stain may be used to adjust for cell-density differences. ATP citrate lyase is the primary cytosolic source of acetyl-CoA and has a central role in de novo lipid, fatty-acid and cholesterol biosynthesis; phosphorylation at serine 454 can regulate its activity, making this phospho-specific assay relevant to studies of lipid metabolism, metabolic disorders and cancer.
