Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative determination of Activating Transcription Factor 5 (ATF5) protein expression in cultured cells. The assay uses an indirect ELISA format in which ATF5 is captured by ATF5-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. It is described as a convenient, lysate-free, high-throughput and sensitive kit suitable for measuring relative ATF5 levels in cultured cells, as well as for evaluating the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on ATF5 expression.

The assay is reactive with Human, Mouse and Rat samples and is intended for use with adherent cells, with a stated dynamic range of > 5000 Cells. Detection is colorimetric, read at 450 nm, and the kit is stored at 4 °C with a stability of 6 months. Two normalisation strategies are included: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density so that absorbance values can be adjusted to cell amounts and plating differences corrected.

According to the manufacturer, ATF5 is a transcriptional activator of the bZIP family that binds the cAMP response element (CRE), a sequence present in many viral and cellular promoters. The target protein is catalogued under UniProt ID Q9Y2D1, with a reported molecular weight of 30,674 Da, and the kit is listed under the Epigenetics and Nuclear Signaling research area.
