Description

A cell-based ELISA kit from Assay Genie designed for the detection of phosphorylated ATF2 at the Thr71 site in cell lysates and tissue homogenates. The assay targets ATF2 (Phospho-Thr71) and is reactive with human, mouse, and rat samples, with adherent cells as the indicated sample type. Detection is achieved colorimetrically at 450 nm, and the kit is configured for high-throughput use as a lysate-free workflow.

The assay uses an indirect ELISA format in which the target is captured by phospho-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step allows cell density to be accounted for after the colorimetric read. The dynamic range supports measurements above 5,000 cells per well.

ATF2 (Activating Transcription Factor 2) is a leucine zipper transcription factor that binds the cAMP-responsive element (CRE) and can form homo- or heterodimers with c-Jun to drive CRE-dependent transcription; it also exhibits histone acetyltransferase activity toward histones H2B and H4 in vitro. Phosphorylation at Thr71 is associated with ATF2 activation, making the site a key readout for stress-response, apoptotic, and survival signalling pathways. The kit is positioned for research into signal transduction and for studying the molecular mechanisms underlying cancer, neurodegenerative disorders, and inflammatory conditions.
