ATF2 (Phospho-Thr691)Colorimetric Cell-Based ELISA Kit

Assay Genie's phospho-specific Cell-Based ELISA Kit is designed for the qualitative detection of phosphorylated ATF2 (Activating Transcription Factor 2) at Thr691 directly in cultured cells, without the need for lysate preparation. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures the target, and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm. Reactivity has been established for human, mouse and rat samples, and the kit is intended for use with adherent cells. The stated dynamic range is "> 5000 Cells."

ATF2 is a member of the leucine zipper family of transcription factors that binds the cAMP-responsive element (CRE), forming either a homodimer or a heterodimer with c-Jun. It also possesses histone acetyltransferase (HAT) activity directed against histones H2B and H4 in vitro. Phosphorylation of ATF2 contributes to the regulation of gene expression, cell growth and survival in response to cellular stress, and its dysregulation has been associated with cancer, inflammatory disorders and neurodegenerative conditions. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalise target absorbance, and a Crystal Violet whole-cell stain is provided so that colorimetric readings can be adjusted to cell density after the substrate step.
