Description

This Cell-Based ELISA kit from Assay Genie is designed for the detection and quantification of ATF2 protein phosphorylated at Ser112 in cultured cells. Using a colorimetric detection method at 450 nm, the assay provides a lysate-free, high-throughput approach to measuring relative ATF2 phosphorylation in adherent cell cultures. It is reactive with human, mouse, and rat samples and is suitable for assessing the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on ATF2 phosphorylation.

The kit employs an indirect ELISA format, in which ATF2 (Phospho-Ser112) is captured by phospho-specific primary antibodies and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. The kit has a dynamic range of > 5000 cells and is supplied in a 2 x 96-Well Microplate format.

ATF2 is a member of the leucine zipper family of transcription factors that binds the cAMP-responsive element (CRE) and can form homodimers or heterodimers with c-Jun to stimulate CRE-dependent transcription. The protein also possesses histone acetyltransferase activity specific for histones H2B and H4. Phosphorylation at Ser112 has been implicated in the modulation of its transcriptional activity, making it a key target for studying cellular responses to stress and disease.
