ATF2 Colorimetric Cell-Based ELISA Kit

A Cell-Based ELISA Kit from Assay Genie provides a sensitive, lysate-free, high-throughput assay for detecting and quantifying ATF2 (Activating Transcription Factor 2) protein expression profiles in cultured cells. The kit uses a colorimetric detection method measured at 450 nm and is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 cells. It is supplied in a 96-well microplate format and is stable at 4°C for 6 months.

The assay employs an indirect ELISA format in which ATF2 is captured by ATF2-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing plating differences to be adjusted.

ATF2 is a member of the leucine zipper family of DNA-binding proteins. It binds to the cAMP-responsive element (CRE), forms a homodimer or heterodimer with c-Jun, and stimulates CRE-dependent transcription. The protein also possesses histone acetyltransferase (HAT) activity, specifically acetylating histones H2B and H4 in vitro, and its gene is located at chromosomal position 2q32.
