ASK1 Colorimetric Cell-Based ELISA Kit

Assay Genie offers a cell-based ELISA kit designed for the qualitative detection of ASK1 protein in cultured cells. The kit uses a colorimetric detection method at 450 nm in a 96-well microplate format and is reactive across human, mouse, and rat samples. It is intended for use with adherent cells with a dynamic range of > 5000 Cells, and provides a lysate-free, high-throughput workflow for measuring relative ASK1 levels and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on ASK1 expression.

Detection relies on an indirect ELISA format: ASK1-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP to catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density for further normalisation of absorbance readings to cell amounts.

The ASK1 target (gene symbol MAP3K5) is a MAP kinase kinase kinase of the STE11 family involved in stress-induced apoptosis and in the activation of the JNK and p38 subgroups of MAP kinases.
