Arrestin 1 Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie is designed for the quantitative measurement of arrestin-1 levels in cell lysates, offering high sensitivity and specificity for reproducible results in research applications. The assay uses a colorimetric detection method at 450 nm, is reactive with human samples, and is intended for adherent cell samples. The assay is a convenient, lysate-free, high-throughput and sensitive format for detecting the Arrestin 1 protein expression profile in cells. It can be used to measure the relative amounts of Arrestin 1 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. Qualitative determination of Arrestin 1 concentration is achieved by an indirect ELISA format, in which Arrestin 1 is captured by Arrestin 1-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme conjugated to the secondary antibody catalyses a colorimetric reaction upon substrate addition.

Two normalization methods are provided to support the qualitative nature of the cell-based ELISA. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and adjusting for plating differences. The target gene is ARRB1 (UniProt ID: P49407), which encodes beta-arrestin-1, a protein that regulates G-protein coupled receptor signalling through receptor desensitization and resensitization.
