Description

From Assay Genie, this in-cell ELISA kit is designed for the precise and quantitative measurement of ARNT (Aryl hydrocarbon receptor nuclear translocator) levels in cell cultures. ARNT is a key transcription factor that regulates genes involved in xenobiotic metabolism, cell growth, and differentiation, making it a valuable target for research into cancer, metabolic disorders, and environmental toxins, particularly within epigenetics and nuclear signaling. The kit is reactive with Human, Mouse, and Rat samples and is intended for use with adherent cells.

The assay employs an indirect ELISA format in which ARNT is captured by ARNT-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme then catalyzes a colorimetric reaction upon substrate addition, measured at 450 nm. This lysate-free, high-throughput approach detects ARNT protein expression profiles in cultured cells and can be used to screen the effects of treatments, inhibitors, or activators on ARNT, with a dynamic range of greater than 5000 cells.

For normalization, the kit includes a monoclonal antibody specific for human GAPDH to serve as an internal positive control. After colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts. The kit is stored at 4°C with 6 months stability.
