Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the detection of APEX1 protein expression in cultured cells. APEX1, also known as APE1, is described as a multifunctional enzyme central to the cellular response to oxidative stress, including DNA repair and redox regulation of transcriptional factors, with dysregulation linked to cancer, neurodegenerative disorders, and cardiovascular diseases.

The assay uses a lysate-free, high-throughput indirect ELISA format. APEX1 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition that is read at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. The kit can be used to measure relative APEX1 amounts in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators.

Reactivity is reported for Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. The format is a 96-Well Microplate, intended for adherent cell samples. Storage is at 4 °C with a stability of 6 Months.
