Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of APAF-1-ALT in cultured cells, as well as in cell lysates and tissue homogenates. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, where APAF-1-ALT is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The kit is lysate-free and intended for high-throughput screening applications, including evaluating the effects of treatments, inhibitors (such as siRNA or chemicals), and activators on APAF-1-ALT expression.

APAF-1-ALT is a key protein in the apoptotic pathway, involved in cell death regulation and immune response, with dysregulation implicated in cancer and autoimmune disorders. The kit is reactive with human samples, including adherent cells, and uses a monoclonal GAPDH antibody as an internal positive control for normalization. A Crystal Violet whole-cell staining method is also included to determine cell density and adjust for plating differences. The dynamic range is greater than 5,000 cells, and the kit is stored at 4°C with a stability of 6 months. It falls within the cell death research area and targets the APAF1 gene (Gene ID: 317, UniProt ID: O14727-6, OMIM: 602233, Unigene: Hs.724521), which encodes apoptotic protease-activating factor 1 (molecular weight 37,976 Da).
