AP-2 Colorimetric Cell-Based ELISA Kit

This Cell-Based ELISA kit from Assay Genie is designed to detect AP-2 protein expression profiles directly in cells using a colorimetric readout at 450 nm. It is a lysate-free, high-throughput and sensitive assay suitable for measuring relative AP-2 levels in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals) or activators have on AP-2.

Detection is achieved by an indirect ELISA format in which AP-2 is captured by AP-2-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the bound HRP catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density, allowing results to be normalised to cell amounts.

The kit is provided in a 96-well microplate format and is intended for use with adherent cells. Reactivity is reported for Human, Mouse and Rat samples, with a stated dynamic range of > 5000 Cells. Storage and stability are given as 4°C / 6 Months. AP-2 is a sequence-specific DNA-binding transcription factor (Gene Symbol: TFAP2A/TFAP2B; Gene ID: 7020; UniProt ID: P05549) whose dysregulation has been linked to diseases including cancer and developmental disorders, making it a valuable biomarker for research.
