Anti-Pembrolizumab (Keytruda®)ADA Qualitative ELISA Kit

Assay Genie’s colorimetric ELISA kit is designed for qualitative antibody screening against pembrolizumab (Keytruda®), determining anti-pembrolizumab antibodies in serum and plasma samples. It is intended for professional use only and for research use only. The required sample volume is “10 µL” and the total assay time is “140 min”. Kit components include a microtiter plate with break-apart strips coated with reactant, ready-to-use reactive and negative controls, assay buffer, peroxidase conjugate, TMB substrate solution, TMB stop solution, wash buffer concentrate and adhesive foil.

This sandwich assay captures antibodies to pembrolizumab from patient serum or plasma in microtiter wells coated with pembrolizumab. After unbound components are washed away, a peroxidase-labelled specific conjugate is added and incubated. Following a second wash, bound enzymatic activity is detected with tetramethylbenzidine (TMB) chromogen-substrate, and the reaction is terminated with an acidic stop solution. The intensity of the reaction colour is directly proportional to the concentration of anti-pembrolizumab antibodies in the sample. Optical density is measured with a photometer at “450/650 nm” after the stop solution is added.
