Anti-Nivolumab (Opdivo®)ADA Qualitative ELISA Kit

This qualitative colorimetric ELISA from Assay Genie is intended for the qualitative screening of anti-nivolumab (Opdivo®) anti-drug antibodies in serum and plasma samples. It works as a sandwich assay: anti-nivolumab antibodies in the sample are captured by nivolumab-coated microtiter wells, unbound components are washed away, and a peroxidase-labelled specific conjugate is added. After a second washing step, tetramethylbenzidine (TMB) chromogen-substrate is added and the reaction is terminated with an acidic stop solution; the intensity of the reaction colour is directly proportional to the concentration of anti-nivolumab antibodies in the sample.

Required sample volume is 10 µL per well, and the stated total assay time is 140 minutes. Colour changes from blue to yellow, and optical density is measured with a photometer at 450/650 nm within 30 minutes after adding the stop solution. The kit includes a nivolumab-coated microtiter plate with break-apart strips, ready-to-use reactive and negative controls, assay buffer, peroxidase conjugate, TMB substrate solution, TMB stop solution, wash buffer concentrate and adhesive foil for covering the plate during incubation.
