Description

The Assay Genie Anti-Bevacizumab (Avastin®) ADA Quantitative ELISA Kit is an enzyme-linked immunosorbent assay for the quantitative determination of antibodies to Bevacizumab in serum and plasma samples. The kit uses a double antigen sandwich format: bevacizumab coated on the microtiter wells captures anti-bevacizumab antibodies from the sample, a Peroxidase-labelled bevacizumab conjugate is added, and bound enzymatic activity is detected using tetramethylbenzidine (TMB) chromogen-substrate. The reaction is terminated with an acidic stop solution, and the intensity of the colour developed is proportional to the concentration of antibodies to bevacizumab in the sample.

The kit is classified as a quantitative biosimilar ELISA for the Human analyte Bevacizumab, with applications in Anti-Cancer and Wet AMD research. Reported performance characteristics include a required sample volume of 10 μL, a total assay time of 140 min, a detection limit of 31 ng/mL, a spike recovery of 85-115%, and a shelf life of 1 year. Detection is colorimetric, measured with a photometer at 450/650 nm. Kit contents include a microtiter plate with break-apart strips, ATB Standards A-E (500; 250; 125; 62; 0 ng/mL), High and Low Level Controls, Assay Buffer, Confirmation Reagent, HRP-Conjugated Probe, TMB Substrate Solution, TMB Stop Solution, and adhesive foil.

The kit is intended for monitoring anti-Bevacizumab antibodies in biological samples and is for research use only. Cited general advantages of Assay Genie's Therapeutic Antibody & Biosimilar ELISA format include simple and inexpensive operation without need for expensive equipment or special handling, high sensitivity and specificity with no significant cross-reactivity or interference with other proteins present in native human serum or other therapeutic immunoglobulins, accurate performance with low intra- and inter-assay variability, and potential cost savings by enabling discontinuation of ineffective biologic treatments.
