Description

From Assay Genie, this Cell-Based ELISA Kit is designed for qualitative measurement of Annexin A6 protein expression in cultured cells using a colorimetric readout at 450 nm. The assay is described as a convenient, lysate-free, high-throughput and sensitive format that operates through an indirect ELISA method: Annexin A6 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyze a colorimetric reaction upon substrate addition. Sample type is adherent cells, and the assay is configured in a 96-Well Microplate format with reactivity against human samples and a dynamic range of > 5000 Cells. Storage is at 4 °C for 6 months.

Because the Cell-Based ELISA is qualitative in nature, two normalization approaches are supplied: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences. The kit can be used to measure relative Annexin A6 levels in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Annexin A6.

The target protein, annexin A6 (gene symbol ANXA6; Gene ID 309; UniProt P08133; molecular weight 75,873 Da), is a calcium-dependent membrane- and phospholipid-binding protein that has been implicated in membrane-related events along exocytotic and endocytotic pathways, including endosome aggregation and vesicle fusion. Abnormal levels of Annexin A6 have been linked in the literature to cancer, cardiovascular diseases, and neurodegenerative disorders, making it a biomarker of interest in studies of cellular function and disease pathogenesis.
