This assay quantifies the DNA-binding activity of the androgen receptor in nuclear and cell lysates, providing a functional readout of AR transcriptional signaling beyond simple expression-level measurement. Nuclear receptors such as AR translocate from the cytoplasm to the nucleus upon ligand binding by testosterone and DHT, bind to androgen response elements (AREs), and regulate downstream gene transcription. By capturing active AR bound to immobilized oligonucleotide probes containing ARE consensus sequences coated on the microplate, Assay Genie's kit delivers a direct measure of receptor activation state in human, mouse, and rat samples. Typical workflows use approximately 10–20 µL of nuclear extract per well, with absorbance read on a standard plate reader. Reagents should be stored refrigerated at 4 °C, and freeze-thaw cycles of working solutions should be minimized to preserve antibody and probe performance for research use only in vitro transcription factor studies.
