Description

Assay Genie's Amyloid beta A4 (Phospho-Thr743/668) Colorimetric Cell-Based ELISA Kit is an In-Cell ELISA designed for the quantification of phosphorylated amyloid beta protein levels in cell lysates. It provides a convenient, lysate-free, high-throughput format for detecting Amyloid beta A4 phosphorylation and expression profiles directly within cultured cells.

The indirect assay captures the target with a phospho-specific primary antibody, while HRP-conjugated secondary antibodies binding the Fc region of the primary antibody catalyse a colourimetric reaction upon substrate addition, read at 450 nm. The kit is reactive with Human, Mouse and Rat samples, with a dynamic range of > 5000 Cells, and is intended for use with adherent cells. Two normalisation methods are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalised to cell amounts.

By measuring phosphorylated Amyloid beta A4 levels, the kit supports research into the mechanisms underlying Alzheimer's disease, in which abnormal amyloid beta accumulation contributes to the formation of toxic plaques in the brain. It can be used both to measure relative amounts of phosphorylated Amyloid beta A4 in cultured cells and to screen for the effects of treatments, inhibitors (e.g., siRNA or chemicals) or activators on Amyloid beta A4 phosphorylation.
