Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative detection of AMPK1 phosphorylated at serine 485 in cultured cells. It works as a lysate-free, high-throughput assay using an indirect ELISA format, in which AMPK1 (Phospho-Ser485) is captured by phospho-specific primary antibodies and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. The signal is read colorimetrically at 450 nm, with a stated dynamic range of > 5000 Cells, and reactivity confirmed for human, mouse, and rat samples. Sample type is adherent cells.

The kit includes a monoclonal anti-GAPDH antibody as an internal positive control for normalizing target absorbance values, together with a Crystal Violet whole-cell staining method that can be used to determine cell density and adjust for plating differences. Researchers can use it to measure the relative amounts of phosphorylated AMPK1 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on AMPK1 phosphorylation.

AMPK1 is the catalytic subunit of AMP-activated protein kinase (AMPK), an energy-sensing kinase that regulates cellular energy metabolism by activating energy-producing pathways and inhibiting energy-consuming processes when intracellular ATP falls. AMPK1 has been implicated in diseases including cancer, diabetes, and cardiovascular disorders, making this assay a useful tool for cell signaling and metabolism research.
