Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the accurate detection of AMPK1 levels in cell lysates and tissue homogenates. It is a convenient, lysate-free, high-throughput and sensitive assay that can detect the AMPK1 protein expression profile in cells, and is suited to measuring the relative amounts of AMPK1 in cultured cells as well as screening for the effects that various treatments, inhibitors (e.g. siRNA or chemicals), or activators have on AMPK1.

The assay uses an indirect ELISA format: AMPK1 is captured by AMPK1-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition that is read at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be further normalised to cell amounts. The kit has a dynamic range of > 5000 Cells, shows reactivity with Human, Mouse and Rat samples, is supplied in a 96-Well Microplate format, and has a stated storage stability of 4°C/6 Months.

AMPK1 (AMP-activated protein kinase 1) is described by Assay Genie as a key regulator of cellular energy metabolism and a crucial factor in maintaining cellular homeostasis. Its dysregulation has been implicated in metabolic disorders including obesity, diabetes and cardiovascular disease, making it a valuable biomarker for research into cellular function and metabolic disease.
