Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the accurate measurement of AMPK Beta1 phosphorylation at Serine 181 in cell lysates. The assay offers high sensitivity and specificity for detecting the target in cultured adherent cells using a colorimetric readout at 450 nm, and is reactive with Human, Mouse, and Rat samples with a dynamic range of > 5000 Cells.

AMPK Beta1 is a regulatory protein that plays a critical role in cellular energy homeostasis by controlling processes such as glucose uptake and fatty acid oxidation. Phosphorylation of AMPK Beta1 at Serine 181 affects its activity, making it an important target for studying metabolic disorders, cancer, and other diseases. The kit uses a lysate-free, indirect ELISA format in which target-specific primary antibodies capture the phosphorylated protein and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition, allowing qualitative determination of AMPK beta1 (Phospho-Ser181) levels.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. The kit is suitable for measuring the relative amounts of phosphorylated AMPK beta1 in cultured cells, as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on AMPK beta1 phosphorylation.
