Description

This colorimetric, cell-based ELISA kit from Assay Genie is designed for the detection of AMPK beta1 protein in cultured cells. It uses an indirect ELISA format in which target-specific primary antibodies capture AMPK beta1, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. The assay is described as lysate-free, high throughput and sensitive, and can be used to measure relative AMPK beta1 levels in adherent cells, as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on AMPK beta1.

The kit has been shown to react with human, mouse and rat samples, with detection carried out colorimetrically at 450 nm and a stated dynamic range of > 5000 cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be normalised to cell amounts. The kit is supplied in a 96-Well Microplate format and is stored at 4 °C with a stated stability of 6 months. The associated gene is PRKAB1 (Gene ID: 5564; UniProt ID: Q9Y478).
