Akt1 (Phospho-Thr308)Fluorometric Cell-Based ELISA Kit

Designed to monitor Akt1 phosphorylation at the Thr308 residue directly in cultured cells, this Phospho Cell-Based ELISA Kit from Assay Genie provides a convenient, lysate-free, high-throughput and sensitive workflow. The assay uses an indirect ELISA format in which phospho-specific primary antibodies capture the target and Dye 1- and Dye 2-conjugated secondary antibodies allow fluorometric readout. It is intended for qualitative determination of Phospho-Akt1 (Thr308) levels and can be used both to measure relative phosphorylation in cultured cells and to screen the effects of various treatments, inhibitors (e.g. siRNA or chemicals) and activators on AKT1/AKT2/AKT3 phosphorylation.

Reactivity covers human, mouse and rat samples, with cells as the required input type. The reported dynamic range is "> 5000 Cells" and the kit is intended to be stored at 4°C for 6 Months. Normalisation of target RFU values is supported by two controls: a monoclonal antibody specific for human GAPDH supplied as an internal positive control, and an antibody against the non-phosphorylated counterpart of Akt1 (Phospho-Thr308) used to normalise the phospho signal. Fluorescent detection requires a plate reader with two channels at Ex/Em 651/667 and 495/521, which is not provided with the kit.
