Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of AKAP5 (A-kinase anchor protein 5) protein levels in cultured cells, offering high sensitivity and specificity for research applications. The kit uses an indirect ELISA format in which AKAP5-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyse a colorimetric reaction. Because the assay is qualitative, two normalisation approaches are supported: a monoclonal anti-human GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining method may be used to determine cell density for adjusting absorbance values to cell amounts.

AKAP5 plays a role in regulating intracellular signalling pathways and protein kinase A (PKA) activity by anchoring PKA to cytoskeletal and/or organelle-associated proteins and directing cAMP signals to specific intracellular effectors. Dysregulation of AKAP5 has been implicated in cancer, cardiovascular disorders, and neurodegenerative conditions. The protein is predominantly expressed in the cerebral cortex and the postsynaptic densities of the forebrain, and to a lesser extent in the adrenal medulla, lung, and anterior pituitary.

The assay has a dynamic range of greater than 5000 cells, uses a colorimetric detection method at 450 nm, and is supplied in a 96-well microplate format for use with adherent cell samples. The kit shows reactivity with human samples and is stored at 4°C with a stability of 6 months.
