Description

This cell-based ELISA kit from Assay Genie is designed for detecting and measuring AKAP10 protein levels in cell lysates and culture supernatants. The assay uses a colorimetric detection method read at 450 nm and is supplied in a 96-well microplate format suitable for high-throughput screening of relative AKAP10 expression in cultured cells, as well as for evaluating the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. The stated dynamic range is > 5000 Cells.

AKAP10, also known as A-Kinase Anchoring Protein 10, is a differentially targeted adaptor/scaffold protein that binds type I and II regulatory subunits of protein kinase A and anchors them to mitochondria or the plasma membrane. It is involved in protein localization, signal transduction, and blood coagulation, and genetic variations in AKAP10 have been associated with susceptibility to sudden cardiac death. The encoded human protein has a molecular weight of 73,818 Da and maps to chromosomal location 17p11.1 (UniProt ID: O43572; Gene ID: 11216). The kit is reactive with human, mouse, and rat samples.

The assay uses an indirect ELISA format in which target-specific primary antibodies are detected by HRP-conjugated secondary antibodies, with signal developed by a colorimetric substrate and read at 450 nm. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit is stored at 4°C and is stable for 6 months.
