AIRE Colorimetric Cell-Based ELISA Kit

Manufactured by Assay Genie, this Colorimetric Cell-Based ELISA Kit enables qualitative measurement of AIRE (Autoimmune regulator) protein expression directly in cultured cells. Using an indirect ELISA format, target AIRE is captured by AIRE-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction read at 450 nm. The assay is validated for Human and Mouse reactivity with a dynamic range of > 5000 Cells per well, supplied in a 96-Well Microplate format suited to high-throughput screening of treatments, inhibitors (such as siRNA or chemicals), or activators on AIRE expression.

The lysate-free workflow includes a monoclonal anti-GAPDH antibody as an internal positive control for normalizing target absorbance, plus Crystal Violet whole-cell staining to determine cell density for further normalization. The kit is stored at 4°C with 6 Months stability.

AIRE is a transcriptional regulator that senses histone H3 modifications and promotes expression of tissue-specific self-antigens in the thymus, a process central to immune self-tolerance. Defects in AIRE cause autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APS1/APECED), an autosomal recessive disorder characterized by chronic mucocutaneous candidiasis, hypoparathyroidism and Addison disease.
