Description

Designed for researchers studying aryl hydrocarbon receptor signaling, the Colorimetric Cell-Based ELISA Kit from Assay Genie provides a convenient, lysate-free, high-throughput method for detecting phosphorylated AhR at Serine 36 in cultured cells. The kit uses an indirect ELISA format in which AhR (Phospho-Ser36) is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies, with a colorimetric readout at 450 nm. It is reactive with Human, Mouse and Rat samples and is suitable for adherent cells, supporting measurement of relative AhR phosphorylation in response to treatments, inhibitors (such as siRNA or chemicals) or activators, as well as screening studies.

To enable comparison across wells, the kit includes a monoclonal anti-human GAPDH antibody as an internal positive control for normalizing target absorbance, alongside a Crystal Violet whole-cell staining method that allows signal to be adjusted for cell density. Detection operates across a dynamic range covering > 5000 cells, and the assay is intended as a qualitative cell-based tool for research into AHR signaling in contexts such as xenobiotic metabolism, immune regulation and disease mechanisms.

Aryl Hydrocarbon Receptor (AHR) is a nuclear receptor and ligand-activated transcription factor. In the cytoplasm it resides in complexes with heat shock proteins; upon ligand binding it translocates to the nucleus, dimerizes with ARNT, and binds xenobiotic response elements to regulate genes including several cytochrome P450 genes. AHR has been linked in research to regulation of regulatory T-cell and T-helper 17 cell differentiation, inflammatory responses and the onset of lung cancer.
