Description

From Assay Genie, this cell-based ELISA kit enables quantitative measurement of AFX (FOXO4) protein directly in cultured cells using a colorimetric method read at 450 nm. The indirect ELISA format relies on AFX-specific primary antibodies and HRP-conjugated secondary antibodies, with a monoclonal anti-GAPDH antibody included as an internal positive control for normalization. A Crystal Violet whole-cell staining step is provided to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted.

The kit is designed to be compatible with a wide range of cell types and is suited to applications such as drug screening, pathway analysis, and toxicity studies in fields including cancer and neurodegenerative disorders. It shows reactivity with human and mouse samples and offers a dynamic range of > 5000 Cells. The assay uses a 96-well microplate format and is stored at 4°C with a stated stability of 6 months.

The target protein AFX corresponds to the FOXO4 gene (UniProt P98177), a forkhead box transcription factor with a reported molecular weight of 47,941 Da. According to the supplied database entries, FOXO4 is involved in processes including cell cycle arrest, insulin receptor signaling, negative regulation of angiogenesis and cell proliferation, and stem cell differentiation, and localizes to the cytoplasm, cytosol, nucleoplasm, and nucleus.
