Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit enables detection of phosphorylation at Ser346 of the beta2 adrenergic receptor in cultured cells. The kit is configured as a lysate-free, high-throughput and sensitive assay using an indirect ELISA format, in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction read at 450 nm.

The assay is intended for use with adherent human cells and has a dynamic range above 5000 cells. It supports measurement of relative ADRB2 phosphorylation in cultured cells and screening of the effects of various treatments, inhibitors (e.g., siRNA or chemicals), or activators on receptor phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step can be used to account for differences in cell density.

The target, ADRB2 (Gene ID: 154; UniProt ID: P07550), is a member of the G protein-coupled receptor superfamily. Beta-adrenergic receptors mediate catecholamine-induced activation of adenylate cyclase through the action of G proteins, and the beta-2-adrenergic receptor binds epinephrine with approximately 30-fold greater affinity than it does norepinephrine.
