Description

The Assay Genie cell-based ELISA kit provides a lysate-free, high-throughput approach for the qualitative measurement of ACVL1 protein expression in cultured adherent cells. Using an indirect ELISA format, the assay captures the target with ACVL1-specific primary antibodies, then employs HRP-conjugated secondary antibodies to catalyse a colorimetric reaction read at 450 nm. The kit is suited for measuring relative ACVL1 levels in cultured cells and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on ACVL1. It shows reactivity with human, mouse and rat samples and has a stated dynamic range of more than 5,000 cells. The kit is supplied in a 96-well microplate format with a stated storage stability of 6 months at 4°C.

For data normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density so that absorbance values can be adjusted to cell amounts.

The ACVL1 target corresponds to the gene ACVRL1 (Gene ID 94; UniProt ID P37023), also referred to as ALK1, a type I cell-surface receptor of the TGF-beta superfamily. Deficiency in this receptor is associated with hereditary haemorrhagic telangiectasia type 2, also known as Rendu-Osler-Weber syndrome 2.
