Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for the precise quantification of actin gamma 2 levels in cell lysates and tissue homogenates. The assay uses an indirect ELISA format in which actin gamma 2 is captured by target-specific primary antibodies; HRP-conjugated secondary antibodies then catalyze a colorimetric reaction upon substrate addition, with absorbance read at 450 nm. The kit is suitable for measuring relative amounts of actin gamma 2 in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators.

Because the cell-based ELISA is qualitative in nature, multiple normalization methods are required. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. After colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalized to cell amounts and compensating for plating differences.

The kit is reactive with human, mouse, and rat samples, has a dynamic range of greater than 5000 cells, and is supplied in a 96-well microplate format. Recommended storage is 4°C for 6 months. Actin gamma 2 (gene symbol ACTG2; UniProt ID P63267; NCBI Gene ID 72; molecular weight 37,083 Da) is a smooth muscle actin involved in cytoskeletal organization, cell motility, and muscle contraction, and its dysregulation has been linked to cancer, muscular dystrophy, and neurodegenerative disorders.
