Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of phosphorylated ACK1 (Activated Cdc42-associated tyrosine kinase 1) at tyrosine 284 in cell lysates and tissue homogenates. The colorimetric assay is read at 450 nm and is reactive with human and mouse samples, with a dynamic range of > 5000 Cells. It is intended for use with adherent cells.

The kit provides a convenient, lysate-free, high-throughput and sensitive assay that uses an indirect ELISA format. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalized to cell amounts. The qualitative assay can measure relative amounts of phosphorylated ACK1 in cultured cells and screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on ACK1 phosphorylation.

Ack1 is a tyrosine kinase that binds Cdc42 in its GTP-bound form and inhibits both the intrinsic and GTPase-activating protein (GAP)-stimulated GTPase activity of Cdc42, playing a critical role in cell signaling pathways involved in cell growth, survival, and motility. Phosphorylation of ACK1 at tyrosine 284 is known to regulate its activity and function, making it a valuable biomarker for research in cancer, neurodegenerative diseases, and other disorders. The target is encoded by the TNK2 gene (Gene ID: 10188, UniProt ID: Q07912).
