Description

From Assay Genie, this cell-based ELISA kit is designed for the accurate detection of ACK1 (also known as Tyrosine-protein kinase TNK2) levels in cell lysates and tissue homogenates. The assay uses a colorimetric detection method measured at 450 nm and is provided in a 96-well microplate format. It is suitable for adherent cells and is reactive with human and mouse samples, with a stated dynamic range of > 5000 Cells and recommended storage at 4°C for 6 months.

Described as a convenient, lysate-free, high-throughput and sensitive assay, the kit follows an indirect ELISA format in which ACK1 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density for further normalization. The kit can be used to measure relative ACK1 levels in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on ACK1 expression.

ACK1, encoded by the TNK2 gene, is a non-receptor tyrosine kinase that plays a role in cell signaling pathways involved in cell growth, survival, and migration. According to the cited UniProt entry, ACK1 binds Cdc42 in its GTP-bound form, and its dysregulation has been implicated in diseases including cancer and neurodegenerative disorders.
