Description

From Assay Genie, this colorimetric cell-based ELISA kit is designed to detect ACC1 (Acetyl-CoA carboxylase 1) protein expression directly in cell cultures. ACC1 is a biotin-containing multifunctional enzyme that catalyses the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. ACC-alpha is the predominant isoform in lipogenic tissues such as liver, adipose and mammary gland, and dysregulation of ACC1 has been linked to metabolic disorders, making it a relevant target for studies in lipid metabolism, metabolic pathways and related disease research.

The assay uses a lysate-free, high-throughput indirect ELISA format on a 96-well microplate, in which ACC1 is captured by ACC1-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colorimetric reaction measured at 450 nm. The kit can be used to measure relative amounts of ACC1 in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on ACC1 expression. It is reactive with human, mouse and rat samples and is intended for use with adherent cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalised to cell number. The dynamic range is stated as greater than 5000 cells. The kit is stored at 4 °C with a stated stability of 6 months.

Reference information for the target protein includes Gene ID 31, UniProt ID Q13085, OMIM 200350, gene symbol ACACA (UniProt entry name ACACA_HUMAN) and chromosomal location of the human ortholog at 17q21.
